Shrinkage-based diagonal Hotelling's tests for high-dimensional small sample size data

چکیده
High-throughput expression profiling techniques bring novel tools and also statistical challenges to genetic research. In addition to detecting differentially expressed genes, testing the significance of gene sets or pathway analysis has been recognized as an equally important problem. Owing to the ‘‘large p small n’’ paradigm, the traditional Hotelling’s T 2 test suffers from the singularity problem and therefore is not valid in this setting. In this paper, we propose a shrinkage-based diagonal Hotelling’s test for both one-sample and two-sample cases. We also suggest several different ways to derive the approximate null distribution under different scenarios of p and n for our proposed shrinkage-based test. Simulation studies show that the proposed method performs comparably to existing competitors when n is moderate or large, but it is better when n is small. In addition, we analyze four gene expression data sets and they demonstrate the advantage of our proposed shrinkage-based diagonal Hotelling’s test. © 2015 Elsevier Inc. All rights reserved.

Improved mean estimation and its application to diagonal discriminant analysis
MOTIVATION High-dimensional data such as microarrays have created new challenges to traditional statistical methods. One such example is on class prediction with high-dimension, low-sample size data. Due to the small sample size, the sample mean estimates are usually unreliable. As a consequence, the performance of the class prediction methods using the sample mean may also be unsatisfactory. T...
متن کاملShrinkage-based diagonal discriminant analysis and its applications in high-dimensional data.
High-dimensional data such as microarrays have brought us new statistical challenges. For example, using a large number of genes to classify samples based on a small number of microarrays remains a difficult problem. Diagonal discriminant analysis, support vector machines, and k-nearest neighbor have been suggested as among the best methods for small sample size situations, but none was found t...
متن کاملShrinkage-based regularization tests for high-dimensional data with application to gene set analysis
Traditionalmultivariate tests such asHotelling’s test orWilk’s test are designed for classical problems, where the number of observations is much larger than the dimension of the variables. For high-dimensional data, however, this assumption cannot be met any longer. In this article, we consider testing problems in high-dimensional MANOVA where the number of variables exceeds the sample size. T...

Background and purpose: By evolving science, knowledge, and technology, we deal with high-dimensional data in which the number of predictors may considerably exceed the sample size. The main problems with high-dimensional data are the estimation of the coefficients and interpretation. For high-dimension problems, classical methods are not reliable because of a large number of predictor variable...
متن کاملA multivariate two-sample mean test for small sample size and missing data.
We develop a new statistic for testing the equality of two multivariate mean vectors. A scaled chi-squared distribution is proposed as an approximating null distribution. Because the test statistic is based on componentwise statistics, it has the advantage over Hotelling's T2 test of being applicable to the case where the dimension of an observation exceeds the number of observations. An appeal...
